چکیده

Introduction: Molecular interactions between glycosaminoglycans (GAGs) affect the behavior of extracellular matrix in health and disease. However, their heterogeneous composition, distribution, and physical-chemical properties complicate their characterization in situ. Here we present a new approach to measuring intermolecular electrostatic repulsive forces between GAGs (and between other matrix macro-molecules), using equilibrium sedimentation (centrifugation) and a new model of macromolecular electrostatic interactions in a centrifugal field. We then demonstrate this method using a well-characterized molecular analog of GA Gs, polymethacrylic acid (PMAA). Theory, Materials and Methods: With a centrifuge, we apply a known, spatially varying mechanical force to a macromolecular solute (e.g., see (!)). From the resulting measured macroscopic solute concentration profile, Ci(r) within the centrifuge cell (Fig!), we calculate a profile of intermolecular spacing . At thermodynamic and mechanical equilibrium, the electrostatic, centrifugal, and diffusive forces, respectively, balance, leading to: Mj(l -vjp)w2r2 RT l () (I) (r) o = 2ziF ziF n Ci r .

Nanomechanics of opposing glycosaminoglycan macromolecules.

In this study, the net intermolecular interaction force between a chondroitin sulfate glycosaminoglycan (GAG)-functionalized probe tip and an opposing GAG-functionalized planar substrate was measured as a function of probe tip-substrate separation distance in aqueous electrolyte solutions using the technique of high resolution force spectroscopy. A range of GAG grafting densities as near as pos...

Interaction between brush layers of bottle-brush polyelectrolytes: molecular dynamics simulations.

Interactions between tethered layers composed of aggrecan (charged bottle-brush) macromolecules are responsible for the molecular origin of cartilage biomechanical behavior. To elucidate the role of the electrostatic forces in interaction between bottle-brush layers, we have performed molecular dynamics simulations of charged and neutral bottle-brush macromolecules tethered to substrates. In th...

Diffusion of particles in the extracellular matrix: the effect of repulsive electrostatic interactions.

Diffusive transport of macromolecules and nanoparticles in charged fibrous media is of interest in many biological applications, including drug delivery and separation processes. Experimental findings have shown that diffusion can be significantly hindered by electrostatic interactions between the diffusing particle and charged components of the extracellular matrix. The implications, however, ...

Compressive nanomechanics of opposing aggrecan macromolecules.

In this study, we have measured the nanoscale compressive interactions between opposing aggrecan macromolecules in near-physiological conditions, in order to elucidate the molecular origins of tissue-level cartilage biomechanical behavior. Aggrecan molecules from fetal bovine epiphyseal cartilage were chemically end-grafted to planar substrates, standard nanosized atomic force microscopy (AFM) ...

Transport and equilibrium uptake of a peptide inhibitor of PACE4 into articular cartilage is dominated by electrostatic interactions.

The availability of therapeutic molecules to targets within cartilage depends on transport through the avascular matrix. We studied equilibrium partitioning and non-equilibrium transport into cartilage of Pf-pep, a 760 Da positively charged peptide inhibitor of the proprotein convertase PACE4. Competitive binding measurements revealed negligible binding of Pf-pep to sites within cartilage. Upta...
